I am working on Plant tissue culture, recently my research plant showed green patches on the white callus and I had doubt that it may be somatic embryos. Therefore, I would like to know, is there any simple technique to visualise these somatic embryos under light microscope.

1) If you scan the tissues under the dissection microscope, you can spot somatic embryos- they will be whitish to pale yellow globular structures or heart shaped or easily separable torpedos.
2) shiny yellow masses on the surface of calli/ brown tissues is easily identified as embryonic masses.
3) if you suspect that a mass is embryonic mount a small speck of tissue on a slide and see under compound microscope. The % of cells that are small isodiametric and cytoplasmically dense indicates that they are embryogenic. I usually estimate the correct PGR concentrations/ removal of salts using 1/2 x or 1/10th salts to induce embryos.
4) if this is your first try, experiment with carrot calli and then it becomes easy to understand the process. I have good experience with mango SEs, Coffee, rose, datepalm, etc. Easiest is to standardize by chopping up a ZE and watching it form Embryogenic tissues. Good Luck
1. It should be good if you can attach an image of them for us to see.
2. Some somatic embryos should be able to been seen under dissection microscope without staining. What plant species you are working on?
