I was planning to perform a cytotoxcity assay wherein I am planning to use the culture supernatant stored frozen at -80 deg C at different time points.
Is there any suggestion with regards to the type of the assay to be used?
LDH assay is there in mind but not sure if it would be effective on stored frozen media which is thawed and used for assay.

The better question is whether the cytotoxic product in the supernatant is stable at -80C? Your experiment is not clearly defined so it is difficult to advise you, but there are several methods commonly used to estimate cytotoxic including LDH release, also MTT, or just simple cell counts to compare relative growth. A google search will easily locate the methods.
