Recently I started working with CHO-Freestyle suspension cells, but our laboratory does not have an orbital shaker. I tought it would be okay without it, so I just used a normal T25 flask. But the majority of my cells are always adhered to the botom of the flask, and sometimes clumped in big chunks (they break up after pipetting). Is there any way to fix these issues without a shaker, or I sould try and get one in the future?
Dear Tamas i think that CHO as Expi293 need oxygenation to growth at high cell density and if you do not have a fermenter i think that you need a shaker and c cup vented Erlenmeyer flasks to achieve this results.
If you cannot buy a CO2 regulated shaker (as new Brunswick S41 or infor multitron) which can cost more than 10k euro, you can buy a standard shaker platform to fit inside a CO2 incubator.

If you are not able to buy a shaker you can also try different way to move the cells as for example the spinner bottles

for which you will just need to insert a magnetic stirrer into the CO2 incubator but sincerely i never used it and i do not know how it perform.
