We are going to count Colony Forming Units of Streptococcus mutans in children 6 to 12 years of age. The salivary samples will be serially diluted from 10-1 to 10-6. We do not want to plate all dilutions so my question is which of these dilutions would be recommended to plate?

I suggest to you to plate all the dilutions (from -1 to -9) at least once. Consider plating these saliva samples for some type of patients (either age classified or caries risk classified). After you do this initial platting, then you'll have a better understanding as to what to expect to be your best dilution to count the cells. Another piece of advice is to use the microdrop plating technique. In our experience this method has given us the lowest coefficient of variation during the statistical analysis in addition to save precious resources and time.
