Dear All,
I try to using ImageJ to merge cell images of red, blue, phase contrast which are RGB image type. According to some instruction, I have to convert them into 8/16 bit before merging them, but I notice that the image quality/resolution would be loss.
Is there a method to just overlay/merge phase contrast and fluorescence images into single image without loosing any resolution?

What type of image do you have now?
You could also just display each individual channel (3x) together with a merged image (ignoring the issue), creating a 4-panel figure that contains as much info/resolution as possible.

the best way to make your overlay is transforming yor RGB image to 8/16/32 bits (more bits, most weight). Then you can asign an apopiated LUT and merge channels. This procedure could altere image bit depth (but if you have an 8 bit RGB color image transforming to 32 bits it shouldnt be a problem), but preserves the resolution. It is important to work with TIF images, if you have formats like JPG it would result in loss of resolution. When you generate fluorescence images in the microscope (not true color) save your images as gray scale (8,12,16,etc depending your camera) in TIF/microscope formate. Then you can convert it in RGB in ImageJ. Or just save both type images.
