My friend sent the short film to me saved as attachment for mp4 format (the file was 1.47 MB). Could someone help me to identify the algae in the short film ( the target algae was the stripped and moving one with a little transparent, not the green tiny circular one)? The sample was taken at the bottom of shrimp culture pond. Thank you!

Hi all, me too had such obervation recently in my samples. I this algae has much aerotopes which is commonly absent Oscillatoria sp.it could be a member of Limnothrix ?.. i am not sure about it but certainly it cant be a member of Oscillatoria.

Solitary trichomes, aerotopes usually present in vegetative stage (they r lacking only in hormogonia and resting stages or under stress), usually without constrictions at cross wall.

It´s a nice film of a cyanobacterium belonging to the order Oscillatoriales, a filamentous non heterocytous group. It´s possible to see the rows of granules on the cross wall, but to be sure about the genus it´s necessary to know the diameter of filaments and the lenght of cells. Célia sant´Anna

It could also be a Beggiatoa sp. The species of this genus live on the top of anaerobic sediments and form white mats. The cells store sulfur inside (granules).
I agree with Célia sant´Anna, Oscillatoriales. For the genus we need more. Maria

OK, Thanks all! It seems difficult to tell the correct specie name by microscopic morphology. If is a condition, molecular identifiction should be carry out to further comfirm.
Zhenfan, Though it can be difficult to ID morphologically - people use morphology as the first step in systematic studies, in counts to verify blooms, for studies on flora, etc. Scientists have been using cyano- morphology since Linnaeus.
I think the reason it is difficult for most (incl me) is like another researcher commented - it is quasi- impossible to get a species name (and for Oscillatoriales a genus) without the diacritical characters that are necessary to ID
I would suggest this book: Süßwasserflora von Mitteleuropa, Bd. 19/2: Cyanoprokaryota: Bd. 2 / Part 2: Oscillatoriales (German Edition) . You can most likely key it out. Also search for local floras of your region
However, molecular data will show you the closest hit in the GenBank Afterwards, it is necessary to align it with other taxa to have more power as to your entitiy, and verify chimera, artifacts, etc etc . Which gene(s) are you planning on using?
If it is not a cyano - then you can also follow the same procedure but just search for non-cyano bacterial studies.
