I am planning to assess hypoxia in colorectal liver metastases. Samples have been snap frozen and stored at -80c. I have done immunohistochemistry for HIF on fixed tissue.
Please could some one advise me on this?
you can detect the HIF-alpha in the tissue extract following protocol
extract the tissue using with Rippa buffer with protease inhibitor and
Sonicate 15 second with ice and Centrifuge at 12,000 rpm for 15 min at 4°C.**
7. Add 4 x buffers in the tube and heat the sample 100 °C for 3min.
8. Vortex and centrifuge 3 min 5000 rpm ready to use gel or store -80°C
** Take small volume of lysate and estimate protein amount

Protease inhibitor cocktail tablets from Roche Diagonastic
Thermo scientific product Cat#89900 Rippa buffer 1x solution (100ml).
Take 10 ml of rippa buffer in 15 ml conical tube and add 1 tablet of
Protease inhibitor vortex & mix until dissolve the tablet.
Aliquot in 1.5 ml microfuge tube (each tube 1ml of rippa buffer) and store at -80°C
When you transfer gel to membrane for Western blot, make sure to do at least 3 h at 60V at 40C sicne HIF-1a is a big protein. Good luck!
