In collecting samples, when my patient's samples type is a tissue, and there is a difficulty in collecting control tissue samples that matches the criteria of my patients can I collect BLOOD samples as controls, knowing that I study the association between some polymorphisms in EGFR gene and lung cancer?? I based my opinion depending on that SNPs are stable, inheritable and don’t differ with cell type?
You are ríght in assuming that SNPs are indeed inherited and stable accross tissues (including blood for that case) so that for instance an association study could be performed comparing lung and blood genotypes. However tumors are more prone to somatic mutations , so your tissue might not faithfully reflect the patients' genetic background. If this is what you are after, you might as well get patient blood instead.
THANKS a lot for your specific answer. I have 100 blood samples but I need another 30 samples to be significant. unfortunately, I can't get this number as a blood, however I found the remaining number as old tissue samples. @Jose Ramon Bilbao
similarly you can use other tissue sample which is tumor free (non tumorous tissue) from the same patients such as frozen tissue and formalin-fixed paraffin-embedded (FFPE) tissue.
