How can I measure MMP-1 protein activity or protein level from liver tissue?

Instead of checking only the mRNA level, I want to check the active protein level of MMP-1 in Liver tissue from mice. How can I do that?

Hi, I think ketamine is the likely cause . You may try inhalation anesthesia with isoflurane. Make sure that the clamp does not occlude the IVC in addition to the portal triad. You may decrease the ischemia time if the mouse liver is steatotic, cirrhotic or cholestatic. Good Luck!!

In addition to the above-mentioned, below are the points for further discussion.

1. Ischemia-reperfusion of 60 min normally does not lead to liver infarction (thus mortality should not be high).

2. How did you realize 70% partial liver ischemia?

3. You may try a suture single detachable knot instead of a clamp.

Sorry, what is the question? I agree with both comments above. 70% resection does not kill the mice so even if all parenkyma is loss they should survive. It may be so that mediators from the reperfused liver do that...
