Kelly, you should be able to do so. Tetramers should not activate T cells as APC or PHA. People routinely used tetramer guided cell sorting for T cell cloning. We used to do that but rarely now as these reagents are not cheap and as we often deal with new specificities. If you sort them into one well, you should provide irradiated feeders to separate these cells to reduce potential by-stander killing. Normally this should not be a problem.
