I performed an ELISA to quantify circulating immune complexes from blood sera using the C1q capture based assay from Quidel. I am interested to learn what the immune complexes are composed off and have access to a mass spectrometer. Is it possible/does anyone have experience of:
a) harvesting bound proteins from an ELISA plate?
b) cleaning/purifying/preparing these so they are fit to be run on a mass spectrometer?

Please be aware that the capacity of an ELISA plate is quite low and hence usually not sufficient for MS. However, under optimized conditions with very sensitve mass spectrometers this approach may be possible. The traditional way is to use immunoprecipitation or affinity chromatography. In this case, you need to have access to the antibodies against your target.
