After subculturing of cancer cells, I place the Petri plate and t-flask into co2 incubator. cells are attached normally but after 24-72 hrs, cells became disappearing. Even under the microscope examination also we could not find any cells and also cells debris. The medium looks a very clear one... Pls, help me with this problem.
and if I placing any one of a flask into neighbors incubator, cells are grown healthy and normal.
I never faced it in my experiments. It can be viral contamination. Clean the incubator and inform your EHS. If you have time to explore the problem: take a small volume of the lysed culture and add to 1-day healthy normal cell culture to see the lysis. Do not forget control plate and use a separate CO2 incubator, which can be a small desiccator with injected CO2. Otherwise, you can damage other experiments.
Thank you for your best suggestion. and whenever i am using MTT plate for cell seeding, cells are growing normally in the same incubator. so wr is dat black sheep??????? i hope you, now u r understood my whole problem... thanks in advance....
