I am planing to differentiate human monocytes into macrophages by M-CSF. While doing flow cytometry, I want to exclude DCs from macrophages to get a pure macrophage population. I am wandering which CD markers can be used to differentiate macrophages from DC? Does anyone have any experiences of expression of CD124, CD54 on DCs? I guess CD54 and CD124 express on macrophages, not on DC.

Hi, Abdullah. I used to differentiate DC from macrophages with CD1a vs CD14. Macrophages will be CD14(hi) CD1a(lo) and DCs CD14(lo) CD1a(hi). Of course, to differentiate DC from monocytes you need GM-CSF and another cytokine, I used IL-4. I don't think you can expect much differentiation into DC by only using M-CSF. Another difference is the timing, we usually used DC after 5-6 days of differentiation.
