Hi friends!  

I've been working with knock-in heterozygous mice with ~10 point mutations in the Cald1 gene (homozygous lethal).  After have trouble for a long time with genotyping, I outsourced the genotyping to two companies and neither could get the allele specific PCR to work...  I was recently recommended to do PCR for the neomycin cassette.  

I've done about 13 new samples and all have come up positive for neomycin (should have been at least some WT in there, right?!  I've been doing a het x het cross)

My question:  Can you use neomycin PCR to distinguish between heterozygous and WT transgenic mice?  Or is it not suitable in some cases?

Attached is my gel from today.  If this is not a good method, I will try to TA clone my gene to see if it is even present in the mice.
