Hi,

I am just starting out with viral transduction and want to transduce primary human blood derived gamma delta T cells.

I have read that gd-T and NK are harder to transduce than ab-T cells and gammaretroviral vectors are better for these then standard viral and phoenix packaging and envelope vector RD114 (Article Efficient Gene Transfer to Hematopoietic Repopulating Cells ...

) is often used and transduction is done after activation (so the cells are cycling). MSCV promoter appears to be superior for transgene expression in primary ab-T cells (but in the Lentiviral setting:

Is the above true? I wasn't able to find a quantitative comparison of lent vs retro and ab-T versus gd-T

Also, could I please get a recommendation for a good empty retroviral transfer vector to use for cloning my transgene?

Some have used the SFG vector (1) SFG vector (https://www.addgene.org/22493/) but I don't see an empty vector.
