Apologies for the probably stupid question but I'm new to the field and forgot to put my glycerol stocks in the freezer, will they be ok over the weekend? Thanks for any help!
You might expect to lose some viability, but I suspect you'll be able to recover colonies upon streaking for isolation. I'd freeze at -80*C when when you get into the lab next. It might depend on what you're doing with these stocks, though. If it's just plasmid storage, and if you have a backup stock of actual plasmid somewhere in the lab I wouldn't worry too much.
Should not be a problem, the glycerol will have minimal effect, so it is essentially the same as putting your culture in the fridge and then preparing a freezer stock a few days later.
