I used DMEM/F12 supplemented with B27 (without VitaminA), 20ng/ml EGF and 20 ng/ml bFGF to culture Mammospheres from patient derived tumor samples or cell lines such as MDA-MB-231 on suspension culture dishes. I cultured microspheres from the MDA-MB-231 cell line using DMEM/F12 supplemented with low concentrations of serum. The serum concentration is about 5%. This needs to be optimized according to the actual situation. You can try to gradually reduce the serum concentration.
