Hi RG community,
I want to use HEK293 FreeStyle cells to express an endogenous edited protein. They are optimized to grow in suspension; however, for clone screening purposes, I would like to have them on plate, like conventional HEK293. Has anybody tried it? Do they work on plate?

Hi, we have done the opposite, conditioned HEK293 (Flp-In) cells to grow in suspension, so I think yes. You can try to reverse the conditioning process detailed here, see page 7:

In short, you could plate the cells in suspension media supplemented with a small amount of DMEM (or whatever you normally use for adherent). Slowly increase the serum % and ultimately reverse the ratio of Freestyle medium to DMEM until finally you are in DMEM with your standard serum %.

Yes this can be done. Just put your cells in T-flasks and turn off the shaking platform or move them to a 37 degree incubator with CO2.

Yes, even in freestyle media. But you can also change to another media if desired.
