I am interested in studying immune cell populations in human tissue. However, the only tissue I am able to get access to is from post-mortems. The tissue may have been "unviable" for approximately 24 hours. Does anyone know if it's still possible to obtain live/viable cells and if any commercial buffers are available to prevent further degradation in downstream processes?
Unfortunately I have no control over the conditions. However, for a significant portion of time the tissue will be kept at 4 degrees. I will be looking at t-cell and b-cell populations and their activation status. I understand that cell surafe marker expression can change when a cell dies. Just looking for advice if anyone has done anything similar and got any tips.
Personally I think that depend on the tissue you want to study, in my case I transport fat tissues at 4 degree (It is only for 4-5 hours in my case. But I know from the bibliography at 4 degrees you can storage it during 24 hours). Also be aware of maybe it is necessary leave the tissue at room temperature for some hours (In my case I can do it directly).
About the cell surface marker yes, cell surface marker can change depending of multiple factors... but also depend on the kind of marker you want to study. However I would recommend you to study cell surface marker of alive cells (For that you can use a live/death fluorochrome such as calcein, sytox red... there are a lot depending of the filters you use) so you make several gates to study your set of markers only in alive cells (anyway, you must keep in mind as you said they can change... they can be apoptotic cells which also you can discriminate with other stains etc.)
