I have read that resetting MS is tricky, so unsure of how to proceed, in 15 years have not had to deal with this as a problem before
Media has set sloppy and wondered if possible to reset. Anyone who could assist with this issue, it would be much appreciated?
Ok, the experiment with adding gelrite went quite wrong. So I have answered my own question. In the back of my mind I remembered something from many years ago with the cation exchange between MS and gelrite. particular with calcium chloride ions.
More than just setting of the gel, I would be concerned about the resultant concentrations of all the components of the medium - the macro-nutrients , the micro-nutrients and the plant growth factors that was added to the medium.
In spite of formulation, gellam gum needs Ca and/or Mg for gellfication.
About resetting, I never tried with culture media gelled with Gelrite or Phytagel however with agar I have observed a decreasing in gel strength when heated again.
Thank you Indu and Ricardo for the return answers. It was one of those accidents which intrigued me in how to fix and why the end result was not fixable.
I think for agar, you can add more agar and re-autoclave it. But, the bad thing is that this prolong high-temperature heating can degrade your nutrients in the medium, such as sucrose.
