I am afraid that some components loaded in Opti-MEM might affect the change in morphology of cells since I won't remove transfecting media out after transfection.
I am thereby considering to use serum-free medium (DMEM/F12) to dilute the DNA and lipofectamine reagents for transfection.
In addition, this experiment, cells are treated with drugs dissolved in serum-free medium.
Is it possible to do ?
Is transfection's efficiency the same between Opti-MEM and serum-free media?
opti-mem simply has certain fractions of purified BSA to feed the cells (to make it "less sticky"). Add some good quality albumin (2-5% final) to your serum-free media and this should work out well.

Yes, you can use serum-free media, if you don't want to use Opti-MEM. We have used serum-free media many times for transfection in a similar scenario like yours. No issue with transfection efficiency.

Yes, it is highly recommended NOT to use any antibiotics during transfection. It affects its efficiency. We never used antibiotics during transfection. Mix your transfection reagents and plasmid in serum-free media, add to cells and go for incubation. It will work.
