We are trying to find if our bactreia strains (Pseudomonas syringae pv. syringae) can produce siderophore that can acquire copper from the media.

We used the classic assay of (Schwyn and Neilands., 1987) but with a difference of using copper chloride (CuCl2 2H2O) instead of iron chloride (FeCl3 6H2O) in the blue cas dye. 1 mM of copper chloride was used with 10 ml of 10 Mm of Hcl ( 0.0017 g of CuCl2 for 10 ml of Hcl) has been used .

The color we got after pouring the media in the plates was first brown-red and then after the media hardened it became transparent-Gray not Blue.

-Can anyone give me some advises? where have I made a mistake?

-Does copper binds with cas the way iron does?
