Can we put the prepared medium containing stimulatory cytokines like MCSF or interleukins at +4C and re-use it again after few days or we should prepare new medium every time?
It is always better to add interleukins in the fresh media just before plating the cells considering the stability of the interleukins/cytokines. However, you can try putting the complete media with cytokines in 4 degrees, but you would see their activity will diminish with time and would hamper the differentiation/culture of cells.
Mohammad Aqdas Alright, That means I have to thaw and freeze the aliquoted cytokines more than 3 - 5 times because I use a small amount of them every time. I am afraid that the cytokines lose its activity when freezing and thawing 5 times.
Ahmed Al-Omar Yes, you should definitely thaw the cytokine/chemokine just before adding and culturing the cells. The 5 times freeze thawing can be avoided too: just make aliquot of the cytokine in the desired concentration (according to your cells/culture conditions) and volume such that you use up all the cytokine and do not re-freeze it.
