I am working on Arabidopsis Thaliana, and also interested in its study related to roots, but i want to know that whether we can measure the diameter of Arabidopsis roots, If yes then please refer me any article or any software that can be useful in this context.
I have tried to measure the diameter with bright field microscope but was not successful , tissue is so delicate to cut accurately..so want to any other alternate approach or any modifications with bright field microscopy

i have not done these experiments but have few suggestions:
1. you may think of staining roots with propidium iodide and take confocal sections. measuring would not be a big problem with the help of manufacturer provided software or free software such as image j. if you think you may squeeze roots while mounting between slide and coverslip, use spacers (arrange 2 more cover slips on either side of your sample before you place cover slip on sample).
2. you can think of fixing samples fixing, embedding in paraffin and taking sections with micro-tome.

Thank you Very much for all your fruitful suggestions, will try them to get the result,
But regarding the second option actually we don't have microtome in our lab so have to find any alternative for cutting.
For your information:
1. There is this company (Regent Instruments Inc.) carrying image analysis system WinRHIZOTM which can be used for plant root diameter measurement (see attachment, http://regent.qc.ca/assets/winrhizo_about.html ). I also attach a paper which contains data and results derived from using WinRHIZOTM system (attachment 2).
2. Another attached paper (2009) reviews available simaging systems for plant root measurement (attachment 3). It was published in 2009, 7 years ago. By now, these systems might have upgraded versions. Just for your reference.
