In one study, we are intending to measure eNOS activity/phosphorylation in tissue specimens. Our study setting fits most with chemical formaldehyde fixation. I was wondering if there is a way to measure enzyme activity in these specimens.

The fixation of the tissue probably destroyed the enzyme activity. You might be able to use immunological methods such as immunohistochemistry to detect the presence of the protein in the sample. Proteomics methods are another possibility for detecting the presence of the enzyme, if the formaldehyde fixation doesn't prevent it.
