I'm studying on marigold oleoresin extract which contain lutein. For TLC, HPLC etc. studies I need standard to confirm its prescence. Sigma standard is available but it's too costly. INR 25,000/mg. Some authors have mentioned the used use of nutracapsules for lutein as their standard in their work. But don't know exactly what they are. Can you please help in getting different materials that can be used as a standard? And how much material do I would like to have for standard.
Since carotenoid/xanthophyll standards are generally pretty costly, commercially available lutein supplements may be a viable option. You may also consider lutein extraction from lutein-rich plant material.
However, compared to the Sigma standard none of the options above are pure and you might run into problems based on the non active ingredients of the supplements or plant matrix. If you don't need a pure standard, then I would get a liquid supplement and try it directly as is.
However, supplements and plants may contain not only lutein but also its isomer zeaxanthin. You can try to isolate the lutein from the matrix and maybe even from zeaxanthin (e.g. prep-HPLC) but you will need to look into the condition to ensure good separation between the 2 isomers. This can be a quite labor intense process and lutein is a fairly unstable and will most likely degrade at least to a certain degree during the process no matter what.
Please note that any compound you purify you should structurally verify by NMR.
So, if you really need the pure lutein standard I strongly recommend the Sigma product. Even though it is costly, it will save you a lot of time, money, and headaches.

I would only add that in leaves carotenoids interact with light their ratio is changing in vivo . This means that it is necessary to couple quenching analysis. Probably dired egge yolk canbe a secondary standard once known the Zeax content as well. http://www.ncbi.nlm.nih.gov/pubmed/24419657
Well, if you really want to quantify you need to use a standard. For that we Always bought lutein from DHI Water and environment (Denmark). Compared to Sigma they were cheaper.

In capsules and supplements are mixtures of carotenoids with major content of lutein for example. So you can not use it for standard

As secondary standard an egg yolk powder is sufficiently free of accessory pigments. What I suggested is to submit a sample to a goverment lab for food safety asking one determination in a certified lab.
