LDH activity in serum was determined kinetically according to Buhl and Jackson.,(1978).

Principle:

pyruvate + NADH+H LDH lactate + NAD+

-the rate of NADH oxidation is proportional to LDH activity.

Calculation:

-an international enzyme unit (U) is defined that activity of enzyme that convert one micromole of substrate in one minute under standard conditions.

Formula:

Enzyme activity(U/L) =

D A / Min.X (Vt x 1000)/(MEC X LP X VS)

=D A / Min x 8200 (at 340 nm and 1 cm light path)

Where

*U/L = international unit per liter.

*Vt= total volume of reaction (ml).

*1000= convert concentration to liter.

*MEC = micromolar extinction coefficient of NADH (6.22)

*LP = light path (cm).

*Vs= sample volume (ml).
