In Plant chemistry, several methods are employed for extraction of crude extract from plant biomass/tissue, concentration of crude extract to contain certain percentage of class or classes of compounds (alkaloids, flavonoids, terpenoids etc.), isolation of classes of compounds from crude extract, isolation of single pure compounds, purification of pure compounds, structural elucidation and identification of compounds. can some one list out the methods under each subheading
1. crude extract extraction from plant material,
2. concentration of crude extract,
3. isolation of classes of compounds,
4. isolation of pure compounds and their purification,
5. structural elucidation of pure compounds,
6. identification of pure compounds.
The selection of solvent system largely depends on the specific nature of the bioactive compound being targeted. Different solvent systems are available to extract the bioactive compound from natural products. The extraction of hydrophilic compounds uses polar solvents such as methanol, ethanol or ethyl-acetate. For extraction of more lipophilic compounds, dichloromethane or a mixture of dichloromethane/methanol in ratio of 1:1 are used. In some instances, extraction with hexane is used to remove chlorophyll

Thank you for your valuable contribution. I checked out the link. I think the methods given there do not completely answer my question which has several mini-questions on the techniques employed from extracting the plant material to isolation and identification of pure compounds.

That's a fairly large question. My response is probably incomplete
1) solvent extraction, Soxhlet extraction, supercritical fluid extraction
2) Evaporation
3) I like to do a column screening using wide polarity range chromatography; others like to do extraction into different solvents;
4) I like to do flash chromatography and prep HPLC chromatography if needed; others do prep TLC
5) Mass spectrometry, NMR, IR, UV-vis spectroscopy. This is supported by a literature search about what was previously purified from the plant. In fact, the literature search may guide the purification, just to verify whether the previously reported compound(s) are the cause of activity in your assays.
