When i prepared primary emulsion (w/o) white layer is precipitated. It may be either PCL or drug. I did not use surfactant for making primary emulsion. My drug is highly water soluble.
dear claudio.... PCl is hydrophobic in nature and my drug is hydrophilic in nature. so in order to prepare nanoparticles, by w/o/w multiple emulsion, drug was dissolved in 1ml of water and emulsified it with 10 ml of dichloromethane using probe sonicator. But I could not get proper emulsion. A thin polymeric film seprated in emulsion. wbat could be the solution for this.
Dear Bhavin, I prepared nanocapsules (w/o/w), with a hydrolifilic drug + iron oxide nanoparticles in a sodium citrate solution + PCL(or PLGA) in dichloromethane + a aqueous solution of Pluronic F-68, this first mixture was sonicated (w/o), then the first emulsion was sonicated with a PVA aqueous solution (w/o/w).
Then, I have in the w1 (drug + iron oxide + sodium citrate aqueous solution + Pluronic aqueous solution)
i tried three solvent for pcl till now. DCM, ACN and , DCM+ACN(1:1). In all cases i am facing same problem. Now, i am going to use ethyl acetate and also method suggested by marcela using poloxamer. thanks to both of you.
@marcela Yes, I have tried DCM and PCL. But the particle size is not good. But PCL and ethyl acetate produced particles with a size less than 100nm. Hence I have worked only with PCL and ethyl acetate.
