I am trying to transfect RNA (especially mRNA) into multiple myeloma cell lines (MM.1S), and I am wondering if anybody can recommend a good reagent that worked for them (i.e kits for transfecting suspension cell lines). I understand that MM.1S cells are difficult to transfect, but I will be happy to get some recommendations. I have tried the Mirus Transit-mRNA kit but it didn't seem to work, and I don't want to keep buying new kits without some data or recommendations that it will work. Thank you! PS: I don't want to use electroporation.

Mirus Transit has been what worked the best for siRNA transfection in our lab. But still, the efficiency was quite low (around 30%) unless we attached the cells (70%), in our case we are using THP1. It may be worth checking if electroporation works for you.
But honestly, what has worked the best in these types of cell lines has been lentiviral transduction after attaching them (80-90%).
