I am working on MAPK pathway in mouse macrophages stimulating by live and killed bacteria. Most of the time I found maximum level of all phosphorylated proteins (p38, JNK, ERK) on WB from all groups control and infected. If some one have experience regarding the detection of these proteins so please, I need help.
Hmmm, a phosphatase inhibitor that is working will lead to maximum levels of P-proteins. If it isn't working you would have more phosphatase activity and less P-protein. Iif you want to decrease your P-protein you need kinase inhibitors. There are separate kinase inhibitors for each distinct MAP kinase cascade.
depending on the system and target, you may have up to 100% dephosphorylation. Another issue would be, is the phosphatase inhibitor cocktail one is using giving 100% inhibition of dephospho rylation for targets of interest.
