I want to prepare a 9 mg/ml solution of RIG012 for mouse experiments. The product manual recommends dissolving the drug in DMSO, with a drug solubility of 33 mg/ml. However, it requires the DMSO concentration to be less than 2%. Are there any good strategies to dissolve the drug?
There are no published reference indicating that RIG012 has oral bioactivity, so oral administration is not recommended. I did find a reference which used i.v. administration. You can have a try.
This compound is quite challenging to dissolve, and no optimal dissolution method has been established. In our labotory tests, a 0.5% CMC/saline solution was able to dissolve it up to 5 mg/mL, which is relatively effective. Although some sedimentation occurs upon standing, it does not cause syringe needle blockage.
