A GeneJet FFPE DNA purification kit was recently accidentally purchased instead of the GeneJet DNA purification kit. I would like to extract DNA from Gram-positive bacterial cells. Provided I lyse the cells using an appropriate solution (20mM Tris-HCl, 2 mM EDTA, 1.2% Triton X-100 and 20mg/ml lysozyme), can the columns and wash buffers from the FFPE kit be used to complete the DNA extraction? The exact make-up of the buffers is proprietary info, I can't be exactly sure if their interchangeable? Anyone here ever tried it?
The analytical chemist in me says you had better check this with the technical support staff at the supplier. I have had trouble with phiX174 closed circular DNA preps when I modified for different reasons. The support staff can probably tell you what you need to do. Good luck, D. Booth
