I wish to study the effect of the monoclonal antibody against a certain protein. Can i purchase this monoclonal antibody for animal study from the widely known companies that are generally used for western blot, IHC etc? If not then why?
I think, generally you purchase only basic research rights for a material. So you can do it as this remains true (check the rights came with the purchase, or discuss it before you order). Technically, you need to remove the preservative, because it is toxic (ie, azide).
If you are asking whether commercial monoclonal antibodies produced against human antigens will cross react with animal antigens, then the answer is maybe. The manufacturer may have species specificity data available that can help with this. If not, then you must test this yourself. Some vendors will provide very small aliquots of antibody for such testing at a nominal price.
If you are asking whether commercial antibodies can be used in an in vivo animal study, the answer once again is maybe. The cost will be very high because relative to in vitro testing large amounts of antibody will be required. Also, the species of origin of the monoclonal antibody (usually mouse) must be the same as the species of animal into which the antibody will be injected.
If you intend to use antibody in-vivo, I would recommend to get antibody prepared on order. There will be multiple issues if you use commercial antibody for this purpose. The vendor may not allow you to get an intellectual property related benefits if the product is successful. The amount of preservative and sometimes vehicle can also be an issue. Also, commercially available antibody will cost you a lot more than the money you need for getting custom made antibody. You can customize almost everything with these custom made antibody at much economic price than commercial one.
Google search will help you find vendors who can custom make antibody and let you have all the rights for antibody.
Animals may develop antidrug antibodies to human antibodies and visa-vera. This may occur a few days following the first dose and compromise PK exposure and effect.
Dear Bret Lum, i was talking about the use of rat monoclonal antibody in the rat model of disease. I guess this will not develop antidrug antibody if so then please explain how.
