We are working with murine BMDMs and while confirming their polarization status by qPCR, we observed that M2 (IL-4) macrophages are expressing IL-1beta at significantly higher level than M0 (unstimulated macrophages). The rest of the profile of IL-4 treated macrophages match that of M2.

Hi Arya, I don't have experience with murine macrophages, but for human macrophages a simple dichotomy might seem logical and comprehensible, but appears rather naive. Using different classic M1 and M2 macrophage markers we found that one third of the macrophages was either double positive or negative (see JAHA paper on the innate immune response in human atherosclerosis). There is quite some literature available on the spectrum of macrophages.
