need to inactivate integrin a2b1.
HUVEC in collagen-I coated well is producing excessive ROS than that of non-coated wells. need to know why... thus need to block the particular integrin signaling. but not sure if this is the right choice. And is using mAbs in solution alright?
need suggestions.
Yes, there are several alpha2beta1 blocking antibodies available, including BHA2.1 and P1E6, that do blunt signaling by alpha2beta1 integrins in endothelial cells. However, you don't know that alpha2beta1 is mediating your effect by just plating cells on collagen. Endothelial cells express other collagen-binding integrins, and the specific repertoire of integrins appears to vary by vascular bed. It would be a better approach to blunt all beta1 integrin signaling using blocking antibodies (ex. AIIB2) or siRNA to verify that integrins are mediating the effect. Then you can follow up with studies on specific heterodimers.
