I am using lentiviruses to transfer my gene into the mES cells and the zeocine resistance gene is present outside of the 5'LTR and 3'LTR region of the transfer plasmid. so to select stably transgene intergrated cell can I use Zeocine antibiotic. Will the plasmid transfer plasmid remains inside the after insertion of gene of insert within LTR into the mES cells genome.
If there is no other antibiotic resistant gene present in the plasmid, using Zeocin may be your only option to select stable cell line.
Shomoita Sayed I used the Zeocine selection to pick the positive clones but my all clones died before revival. I think no other antibiotic site is present with the LTR site of my plasmid. I have left with only one option select clones without selection.
You may try sequencing the clones to get the absolute validation about them then.
