Halo mbak, sebelumnya saya pernah melakukan cytotoxicity test pada sampel cairan, tapi tergantung konteks penelitiannya dulu. Apa bisa dijelaskan garis besar penelitian yg terkait dengan tes tersebut?

The protocol you read in the paper is good enough. You need to pass your supernatant through 0.45 uM filter to make sure of sterility. Regarding the concentration, you can not use u/ml for liquids. I would suggest following dilution factor like 1:10 or 1:1000 etc.

I'm afraid that after centrifugation you are not sure what is remaining in your supernatant sample (unless you are going to analyze it again to determine the resident compounds in it). If the sample you are testing at the beginning was concentration x, that concentration will probably be changed after centrifugation and passage through filter.

You need to find some other way to prepare sample for testing, besides this one,If the sample is in a liquid form try finding another way to sterilize it. If the sample is plant extract (presumably) try to evaporate the solvent you extracted plant material with, or try lyophilisation if the water is in question.

1. You can easily use the liquid form as it is, especially for short-term cytotoxicity assays (I have exposed cells to compounds in liquid form myself up to 72h without any problems. 

2. You can use the density of the compound (in mg/ml) to calculate how much ml you will need to get the concentration you want in micrograms/ml.
