Microscope is upright microscope, and the sample is microfluidic PDMS chip. I would like to have the chip submerged under water so there's a high chance that objective might be submerged in water like water-immersion objective. I'm not sure if it will break down the objective due to any leakage of water into the objective, of if it should be fine.
As Karl has noted, spec water immersion objectives are just that. They are designed to be immersed in water so if you use one in water, then you will be ok. Do not use an air objective in water (or any other liquid).
Yes, I am aware that the regular air objective will cause more spatial deflection if immersed in water compared to that of water immersion objective since it was not designed for that purpose. However, my concern was more about if objective will not break due to exposure to the water. Is objective typically tightly assembled enough to prevent leakage?
No, the adhesives used inside the objective to secure the lenses will leak and destroy or damage the objective. The coatings applied to the lens may also be damaged too.
