I wan to pull out anti-5mC IgG (raised in Mouse, monoclonal) [Antibody Clone-33D3] . Literature suggest using Anti-mouse Dynabeads, which we don't have! Can I proceed with Protein-G Dynabeads?
You can use protein g beads for purification. Protein G has good affinity for mouse IgG. It should do the trick.
Yes you can but if you are using mouse serum or ascites as your MAb source,, you will pull out the normal IgG as well. If the MAb was grown in culture containing FCS you will pull out the bovine IgG as well. (We run our FCS over Seph-protein G before we use it to grow MAbs so we don't have this problem).
Just be aware of this so you know what will be in your final preparation....sometimes it matters and sometimes not.
Thank you for your answers. We have purchased this antibody from commercial source (link given), which says purified antibody. So it should not have normal IgG from FCS or mouse serum. Am I right?
