Hi all, 

i'm thinking in taking advantage of inteins to produce a specific peptide. I'll clone my peptide as the C-extein of the construct.  However, i've a doubt regarding the first amino acid of the C-extein. I've read and quoting "The first amino acid of the C-extein is invariably a Ser, Thr or Cys, but the residues that precede the intein at its N-terminus (i.e. the N-splicing domain) are not conserved". My peptide starts with a Gly and i'm wondering if the activation of the intein will be efficient or not. 

What do you think? If i start the C-extein with a Gly residue the system will work and i'll obtain my peptide?
