I can answer your first question and partly the second - yes, you can use both primers in the same cDNA synthesis reaction as evidenced by kits that are on the market e. g. Maxima 1st strand synthesis kit by ThermoScientific.
As for the concentration/amount... we used to use 20 pmol oligo d(T) primers (10µM stock) with 40 nmol dNTPs (2nM stock) for up to 5 µg RNA in cDNA synthesis so that could be a starting point for you.
you can use both primers together, i have use OligodT with random nanomers.Use higher concn for random primers than OligoDT and low temp for cDNA synthesis (45 C)
You can certainly use both random hexamers and oligo dT primers in a single reaction. Use in 3:1 concentration for random hexamer & oligo dT respectively.
