I want to isolate a full sequence of a HKT (High affinity K+ Transporter) gene from unknown plant species.
According to some papers, Xenopus oocytes were used to confirm the gene but I don't know how to use it and I don't have the tools to analyze it so that I want to try yeast transformation. In the paper, a yeast trk1 trk2 mutant that is
defective for Na+ and K+ uptake was used to transformation but I can't search this strain and I don't know where and how to obtain it.
Therefore, I am planning to use a commercial yeast transformation kit such as S. c. EasyComp Transformation Kit (Invitrogen K5050-01), instead of the yeast mutant. Then, I will show the yeast transformed with the vector including target gene can survive at higher salt concentration than the yeast transformed with the empty vector.
Is it make sense or not? Is it enough to publish a SCI paper?
