Do you guys have any experience in transferring spheroid produced in low attachment plate to a high attachment plate after spheroid production? Would it affect the morphology of the spheroid?
I wish to use the low attachment plate for production of more spheroid. I am not sure about washing it (ethanol? ultrasound?) since it will wear off the coating and stuff. However, is it possible to transfer the spheroid to another cheaper plate (e.g. normal tissue culture treated 96 well plate) for other assay like drug treatment and MTT? Will it produce any inconsistency?

Depending on a particular cell line, spheroids can tend to disassemble and cells spread like grown in a monolayer. Well, for HeLa spheroids 24 h is more or less ok, so, if your experiment lasts not longer than one overnight, it might be suitable. Using low adhesion dishes or layering agarose on bottom of a dish might be an option that could help if you want spheroids to be more spheroidish.
It depends on the cell type used and the time used. If kept for long the spheroids will deform and the cell properties will change over time. Hanging drop cultures or using low attachment dishes can be the options.
