I would like to fix OR prepare activated sludge samples for scanning electron microscopy (SEM).
I would like to replace Glutaraldehyde (GA) but i am unaware of concequences.
Hello,
Glutaraldehyde is the best but you can use paraformaldehyde.
Avoid ethanol, acetone. They fix by precipitation, so you may have deformation of your soft sample.
Paraformaldehyde is preferably use for immunofluorescence experiments with a short fixation time (15 minutes) to allow immunolabelling.
For SEM you can fix your samples much longer (hours, days).
Nicolas Barois Thank you and appreciated.
