Hi all, please give me a hand. I am going to use glibenclamide (from Sigma) to treat diabetic rats as a positive control for my extract. I would like to know which solvent is suitable for dissolving glibenclamide before oral administration. I searched some papers and they use DMSO, however I don't know the concentration of DMSO. May I use the absolute one for rat feed? Or can I use another solvent (which, ?%). Thank you so much!
DMSO is very toxic. if your compound is lipophilic (not solubale in water) the you can disolve it in DMS initially (a concentrated form) then reconstitute in water for administration to animals. However, you have to bear in mind that your control animals would need same diluted, reconstituted amounts of DMSO, hence try to have as lower concentration (volume) of DMSO as possibly to be used. Hope this is helpful. An laternative method, would be disolving your compound in naturally occuring solvents such as olive oil. Let me know if ti works. Good luck.
Yes, of course DMSO is carcinogenic. So, I must say, you should avoid DMSO. You can simply use Water to make solution of glibenclamide. In our experiment, we usually used water or oil.

You can use either of the following:
Water
Oil (peanut)
Oraplus ( ideal as it contains suspending agent too)
but NOT DMSO.
For the compounds what have very pure solubility in water, you can prepare stock in DMSO and dilute in water before administration.
However, please, consider that in cell culture cell can survive even 1% of DMSO (final concentration) and DMSO does not have effect on cell development at the 0,1- 0,2%.
dmso is carcinogenic and cryoprotectant. On the other hand its very good for solubility.ı propose you not to use it.

Thanks all for very helpful and intelligent advices.I tried dissolve glibenclamide in DMSO and dilute in water, the precipitate occurred. however when I dilute in ethanol 5% the solution had milk color with very small bead- like. I am wondering that can it work?
perhaps dissolve in DMSO and then in oil ( cremaphore 10%) , perhaps that will work
