I found that most of differentiating cells (3T3-L1 preadipocytes, HaCaT keratinocytes, C2C12 myocytes....) show very active metabolism and make "the medium color" red to yellow (presumably by acid metabolites) very fast compared to undifferentiated cells or already differentiated cells do.
Now, I attempt to test cheap and fast method for screening agent which promote cell differentiation.
I designed two experiments,
If a specific agent has ability to promote cell differentiation,
1) In the early phase of differentiation, the medium color of cells exposed to the agent would be more yellowish as much as it can be differentiated by spectrophotometer (because the agent makes the cells differentiating step more fast) compared to the that of control cells.
2) In the late phase of differentiation, the medium color of cells exposed to the agent would be less yellowish (because the agent makes the cell already finish the differentiation) compared to the that of still differentiating cells.
Is it reasonable idea? or do you know any articles on this idea that "medium color could be the effective differentiation marker" ?
