For cellulose affinity chromatography-based specific isolation of dsRNA, researchers use CF-11 cellulose powder. CF-11 cellulose, in the presence of 15-16% enthanol concentration, specifically binds dsRNA elements. I am just wondering whether CF-11 cellulose powder needs to be autoclaved before being used for chromatography purposes (as it will be used for RNA work). Does autoclaving have any adverse effect on CF-11 structure/integrity? Please let me know.
There may be an alternative to autoclave: to suspend cellulose powder in large excess of DEPC-treated water and remove the water before use. Further, autoclaving under a controlled condition (temperature reaching only 100 degree C) may reduce any remaining RNase activity.
For extracting dsRNA, you do not need to autoclave CF-11 (dsRNAs are not as sensitive as ssRNAs). Just add the CF-11 to chromatography column.
Whatman CF 11 cellulose powder (Cat. No. 4021-050) has been used by several researchers in the past to separate dsRNA elements from other nucleic acids. CF-11 cellulose under 15-16% ethanol concentration binds only dsRNA fraction. Unfortunately, the producer of CF 11, Whatman, does no longer produce it. Does anyone has experience with alternatives? what is the alternatives to CF-11 cellulose for dsRNA-binding purpose? Please let me know
