I would like to detect signs of oxidative stress in frozen human T Cells of a disease cohort that I collected from patients over the last 2 years compared to also frozen healthy samples. I already read that it is recommended to use oxidative stress detection kits like CellROX and MitoSOX on fresh cells since freezing and thawing might alter the assays´ result. But does anyone have any experience on how to optimize the protocol in order to make the assay work better for previously frozen cells? Maybe by culturing and/or activating the T Cells beforehand?
