I undergo Soxhlet extraction by methanol to test antimicrobial activity of Jade plant(crassula ovata) by agar well diffusion technique but after extraction the extract remain in thick liquid phase, Even if it remains that way I dissolve it in to more liquid state by adding 1ml of distilled water rather than using DMSO the experiment already work out but am still have ambiguity about its phase after extraction because I am thinking if I used DMSO for dissolution it might give it greater inhibition zone since it can able diffuse in the media what do u think what type of technique should I use and repeat it
All extractions are equilibria, which means some compound is always left behind. By changing the solvent, you have changed the equilibrium costant and may have left more of the compound behind. There is also an effect of the cell walls and membranes when extracting from living tissue. The solvent can have an effect on the cell walls and membranes making them more or less permeable to the compound. This latter issue usually only effects the rate of compound extraction, not the equilibrium.
